چکیده

Three juvenile hormone analogs were found to be in vitro inhibitors of rat liver microsomal oxidases. Aniline hydroxylase reactions gave apparent inhibition constants of 8 X 10-5 M and 1.4 X 10 4 M for ( E ) -4[( 6,7 -epoxy-3,7 dimethyl2monenyl)oxy] 1,2(methylenedioxy) benzene and (E) 4 [(6,7 epoxy 3 ethyl 7 methyl 2 noneny1)oxyl 1,2 (methylenedioxy)benzene, respectively. (E)-6,7-Epoxy-l-(p-ethylphenoxy)-3,7dimethyl-2-octene, the least active, had an apparent inhibition constant of 2.5 X M. Cytochrome P-450 binding for all three compounds was observed to be of the type I category. The results provide evidence that J H analogs react with mammalian tissues and are thus not completely insect specific.

Differential Metabolic Effects of Novel Cilostamide Analogs, Methyl Carbostiryl Derivatives, on Mouse and Hyperglycemic Rat

Objective(s) PDE3 has a functional role in insulin secretion and action. We investigated the metabolic effects of new synthetic PDE3 inhibitors (mc1, mc2, mc5 and mc6), on mice and hyperglycemic rat. Materials and Methods The test compound or solvent was injected subcutaneously to mice, for 7 days. On day 8, blood and liver samples were obtained. In hyperglycemic rat, 0.5 g/kg glucose with o...

Identification of Intracellular Sources Responsible for Endogenous Reactive Oxygen Species Formation

The endogenous reactive oxygen species ("ROS") formation is associated with many pathologic states such as inflammatory diseases, neurodegenerative diseases, brain and heart ischemic injuries, cancer, and aging. The purpose of this study was to investigate the endogenous sources for "ROS" formation in intact isolated rat hepatocytes, in particular, peroxisomal oxidases, monoamine oxidase, xanth...

Vitamin A compounds and analogs as inhibitors of mixed-function oxidases that metabolize carcinogenic polycyclic hydrocarbons and other compounds.

Fourteen vitamin A compounds and analogs have been evaluated as inhibitors of microsomal mixed-function oxi dases from liver and lung tissues of mice and hamsters. With liver microsomes, benzo(a)pyrene (BP) is metabolized, by one pathway, to a product (BP-X) which is tightly bound to reaction components. The Km for BP in this reaction is 10 ± 2 /xM.The formation of BP-X is strongly inhibited b...

Chemical structure, reactivity, and carcinogenicity of halohydrocarbons.

This review summarizes studies concerning the covalent binding of [14C]TCE to rat liver microsomal protein and exogenous DNA, in vitro, the enhancement of this binding by inducers of mixed-function oxidases, and inhibition of binding by inhibitors of these enzymes. Furthermore, recent studies on this type of binding in various strains of mice and rats of both sexes and using microsomal preparat...
